The Impact and Change of IL-1and TNF-α on Repair of Cartilage Damage That the Rabbit Knee Joint Were Immobilized

Objective: To observe the change of the levels of IL-1and TNF-α in the synovial fluid of repair of the rabbit cartilage damage, and to discuss the impact of IL-1and TNF-α on repair of cartilage damage that the rabbit knee joint were immobilized。To provide the theory basis for clinical treatment and reduce early cartilage damage in the joint brake Methods.

Group of Experiments: All animals were randomly divided into five groups, immobilizing four weeks (A), after immobilizing four weeks to exercise two weeks (B), after immobilizing four weeks to exercise four weeks (C), after immobilizing four weeks to exercise six weeks (D), the control group(E).

Test method: By the ELISA method to detect the levels of IL-1and TNF-a in A, B, C, D, E five synovial fluid of knee joint.2.

In the state of anesthesia,with the femoral epicondyle articular cartilage of the rabbit side placed into10%formaldehyde to fix in one min, series dehydration, taking off calcium, conventional paraffinning, HE dying, To observe each group the change of each layer of cartilage in the Outlines of the specimen observation and light-microscopy.

Statistical method: The experiment data is analysised by statistical software SP SS13.0. the experimental data is showed by mean±standard.Single factor an alysis of variance is analysised many differences between the group.the two gr oups compares adopt LSD-t method and Bonferroni method inspection.

The co ncentration of IL-1and TNF-α adopts Pearson correlation related inspection. P<0.05shows inspection differences are statistically

Significant Results: Compared with group E, the levels of IL-1in synovial fluid of group A、B、 C、D were significantly differente (P<0.05), the two groups compares of the levels of IL-1in synovial fluid of group A、B、C、D were significantly differente (P<0.05).2.

Compared with group E, the levels of TNF-α in synovial fluid of group A、B、C、D were significantly differente (P<0.05), the two groups compares of the levels of TNF-a in synovial fluid of group A、B、C、D were significantly differente (P<0.05).3.The levels correlation of IL-1and TNF-a in synovial fluid of group A、B、 C、D is closely positive related.

Pearson correlation coefficient is0.953, and is statistically significant (P<0.05).

Conclusion: The levels correlation of IL-1and TNF-α in synovial fluid of experimental animal is closely positive related, in cartilage injury recovery the levels of both is decreased.2. After the termination of the joint braking, free activities can alleviate the cartilage damage, and along with the activity is prolonged, cartilage damage gradually ease.
