Title Study on Extraction Method and Hypolipidemic Effect of Moringa Oleifera Seed Oil
Abstract

Moringa oil(MO) is a vegetable oil rich in mono-unsaturated fatty acids.It is extracted from Moringa oleifera seeds,in which crude fat content are as high as 20-45%.Oleic acid is the largest components in its fatty acid contents.It is about 67-76%.In abroad,Moringa oil is widely used in cooking,cosmetics,machinery lubrication,soap and perfume industries.Moringa oleifera was introduced into China and widely cultivated in recent years.As a source of vegetable oil, Moringa oleifera is a valuable economic plant.Its oil resembles olive oil closely. Study on Moringa oil’s nutrition and Health benefits will surely promote and also help to utilizing Moringa resources.Objectives:Use Moringa oleifera seeds collected from Yuanjiang county, Yunnan province,as study material.(1) To obtain Moringa oleifera seeds oil’s idea extraction method;(2) To identify MO’s fatty acid content、study its characterister of antioxidation stability,and predict MO’s shelf life;(3) And to study MO’s effect on blood lipids profile of hyperlipemic rat model.Method:(1) Determine Moringa oleifera seed’s oil content by using soxhlet extraction method;use orthogonal experiment to study extraction parameters of Ultrasound-Associated Extraction method,Rapid Solvent Extraction method, Microwave-Associated Extraction method,and Aqueous Enzymatic Extraction method;and select out MO’s idea extraction method by compare oil yields of above extraction method;(2) Determine MO’s fatty acid profile by using gas chromatography;and study MO’s antioxidation stability by using Schaal accelerated oxidation method;(3) Build hyperlipemic SD rat model by giving male adult SD rats high fat diet for 7 weeks;according to TC level,divide hyperlipemic SD rat model into positive control group,hyperlipemic control group,MO low dose group,MO medium dose group and MO high dose group by random stratified sampling method.There were six groups,plusing blank control group.Blank control group and hyperlipemic control group are given edible blend oil.Positive control group gives 0.04g/Kg.bw fenofibrate.MO low dose group,MO medium dose group and MO high dose group give 0.5g/Kg.bw, 1.0g/Kg.bw,and 2.0g/Kg.bw Moringa oil,respectively.After 4 weeks’ intragastric administration,determine TC,TG,HDL-C,Organ/Body Weight Ratio and liver histopathological indices.Judge MO’s hypolipidemic effect by comparing MO groups with hyperlipemic control group,Results:(1) Moringa seed’s oil content is 37.2%.The ideal extraction solvent is petroleum ether.Ultrasound-Associated Extraction method,Rapid Solvent Extraction method,Microwave-Associated Extraction method,and Aqueous Enzymatic Extraction method both can be used to extract MO.Ideal extraction method for Moringa oil is Ultrasound-Associated Extraction method.Its optimized parameters are as follows:frequency 28 KHz,ratio of lipuid to material 8:1(V/m,ml/g),extraction times 2,and extraction time 40min.In that condition,the oil yield is 35.85%,and its recovery rate is 96.4%.(2) Moringa oil is rich in mono-unsaturated fatty acids and saturated fatty acids.Their percentages are 74.5%and 25%respectively.Polyunsaturated fatty acid content is relatively small,accounting for only 0.5%.In fatty acid compositions,oleic acid(C18:1) accounts for 70.5%,while Behennic acid(C22:0),palmitic acid (C16:0),stearic acid(C18:0),and arachidic acid(C20:0) stand for 8.61%, 5.65%,4.99%,and 3.76%respectively.Moringa oil contains mono-unsaturated fatty acids and saturated fatty acids mainly,and the oil is also rich in antioxidants. Therefor Moringa oil is stable to thermal oxidation.Without adding any antioxidants,Moringa oil can be stored for about 400 days at room temperature. Moringa oil is unstable to light,so it should avoid exposing to sunlight.(3) Study on Moringa oil’s hypolipidemic effect in SD rats shows that giving an oral dose of 2.0 g / Kg.bw for 4 weeks can decrease blood triglyceride level by 24%and increase HDL-C level by 23%.It suggests that Moringa oil has hypolipidemic effect on blood triglyceride level.Conclusions:(1) Moringa seed’s oil content is 37.2%.Ideal extraction method for Moringa oil is Ultrasound-Associated Extraction method.(2) Main fatty acid in MO is Oleic acid.Therefor Moringa oil is stable to thermal oxidation.Without adding any antioxidants,Moringa oil can be stored for about 400 days at room temperature.(3) Moringa oil has many bioactive substances towards Cardiovascular diseases.Moringa oil has hypolipidemic effect on rats.
