Title The Study on the Neuronal Plasticity in the Auditory Pathway of the Tinnitus Rats Induced by Sodium Salicylate
Abstract

Objective: we discussed the theory of centralization by detecting the expression of Arc、GAP43、egr-1 in the cochlear nucleus、superior olive、colliculus and auditory cortex in the tinnitus rats induced by sodium salicylate.Methods: 1、Established the rat model of tinnitus; 2、Studied the changes of ABR induced by salicylate injection at different time points during making the rat model of tinnitus;3、SYBR Green1 realtime relative quantitative RT-PCR was used to detect the expression of gene Arc、GAP43 and egr-1 in these four auditory nucleis of the rats with early and persistent tinnitus.Results:1、The conditioned reflex of the GroupⅠandⅣtook a longer time than groupⅡandⅤ, and there was significant difference in them(P <0.01).2、There was significantly increased in auditory thresholds of ABR(P<0.01), prolonged latencies of 70 dB SPL waveⅡon tinnitus rats rose with the times of the injection of sodium salicylate(P<0.01).3、Expression levels of Arc mRNA up-regulated in the auditory cortex (P<0.05);Arc mRNA in the cochlear nucleus first down-regulated( P<0.05)then climbed back slightly(P<0.05); Arc mRNA of the colliculus inferior increased and maintained a high degree ,no longer climbe(P<0.01); we failed to find any change in nuclei olivaris superior(P>0.05). GAP43 and egr-1 mRNA in the auditory cortex remarkablely increased (P< 0.01);GAP43 mRNA in the cochlear nucleus first down- regulated(P< 0.01),then climbed back slightly(P<0.01)and egr-1 mRNA declined dramatically(P<0.01); up-regulation of GAP43 and egr-1 mRNA was observed in the colliculus inferior (P<0.01); in nuclei olivaris , we found GAP43 mRNA of groupⅣdecreased(P<0.01) and egr-1 mRNA reduced with increasing injection times of the sodium salicylate(P <0.05).Conclusion:1、The behavioral experiment confirms the rats experienced tinnitus after salicylate injection.2、The ABR testing provid objective audiologic evidence for the toxic effect on ear induced by high-dose salicylate.3、The variation of Arc/arg3.1、GAP43 and egr-1 expression not only support that a shaped changes in the auditory pathway in the rats with tinnitus induced by sodium salicylate ,but also showed that these changes can maintain the long-term tinnitus.
