| Abstract | ObjectiveObservation ofαB-crystallin protein’s role in survival of retinal ganglion cells and the expression of apoptosis protein Caspase-3 in rats after acute ocular hypertension.MethodsSprague-Dawley(SD) rats were randomized into four groups(forty in each group)(1) A group ,acute high intraocular pressure group.(2) B group, acute high intraocular pressure following vitreous puncture group.(3) C group, Acute high intraocular pressure following intravitreal injection of normal saline group.(4)D group,acute high intraocular pressure following intravitreal injection ofαB crystallin protein group. Observing the difference of the survival of rat retinal ganglion cells and the expression of Caspase-3 in each group at different time points after acute ocular hypertension,using of Counting fluorescent gold labeling and immunofluorescence staining.ResultsAfter acute ocular hypertension, the retinal ganglion cells of the animals’right eyes begin to reduce, retinal ganglion cells were all decreased seen under low magnification fluorescence microscopy , in the first week decreased the most, two weeks later, the reduction slowing down. Compared with the other 3 groups, 1 week, 2 weeks, 3 weeks, 4 weeks after acute ocular hypertension, the survival rate of retinal ganglion cells significantly improved, the difference was statistically significant (p<0.01), between the other 3 groups at each time point , the difference was not statistically significant (p>0.05); After acute ocular hypertension, the animals’right eyes appeared positive immunofluorescence in Caspase-3 test,Compared with the other 3 groups, 24 hours, 48hours,72 hours, 1week after acute ocular hypertension, D group significantly reduced the expression of Caspase-3, the difference was statistically significant (p<0.01), between the other 3 groups at each time point , the difference was not statistically significant (p>0.05).ConclusionsαB-crystallin protein has the effection of protecting retinal ganglion cells after acute high intraocular pressure, it could increase the survival rate of ganglion cells.αB-crystallin protein can inhibit the expression of apoptosis protein Caspase-3 in the retinal ganglion cells after acute high intraocular pressure then reduce apoptosis. |
