| Title | Mesenchymal Stem Cells in the Treatment of Perimenopausal Syndrome in Rats |
| Abstract | Objective:To study the effect of BMSCs on perimenopausal syndrome in rats.Methods:BMSCs were isolated from rats. Later, the BMSCs were cultured, purified and identified. There were four groups in vivo experiment:control group, BMSCs group, estrogen group and youth group. The BMSCs group rats were injected 1~2×106 BMSCs, the control group were injected 2 ml saline, the estrogen group were fed estradiol according to 0.158 mg/(kg.d) last for 8 weeks when these rats were perimenopausal period. The youth rats were injected 2 ml saline the same to the control ones. Weighing the rat body weight, ovary, adrenal gland and uterus, the organ coefficient was calculated. Follicle count was finished under the light microscope, and statistics was analyzed between primordial follicles, primary follicles, antral follicles and corpus luteum number. The concentration of estradiol and progesterone were detected with electric light, and detection of AMH with ELSA kit. Visceral index, follicular count, E2, FSH and AMH were compared in each group.Results:1. Organ coefficient:control group (0.1379±0.0508) and MSCs group (0.21±0.0389) were compared with statistically significant (P<0,05). Adrenal and uterine organ coefficient was not significant (P> 0.05).2. Follicle count:In the primordial follicles, primary follicles and secondary follicles, MSCs group is higher than in the control group, but not statistically significant.3. Estradiol E2:after 3M the control group (35.16+3.54pg/ul) and MSC group(49.07±15.00 pg/ul), MSC group had an increasing trend, but not statistically significant.4. Progesterone:after 3M, the control group (3.77±1.43 ng/ul), MSC group (6.29±7.13ng/ul), MSC group had an increasing trend, but not statistically significant.5. AMH test results:intervention 1M, control group (0.924±0.400 ng/ul), MSC group (1.57±0.426 ng/ul) were compared with two groups(P=0.036), after the intervention of 3M, the control group (0.557±0.314 ng/ul) and MSC group (1.265±0.372ng/ul) were compared two groups (P=0.034), after the intervention 1M and 3M, MSC group were higher than the control group, lower than the young group, and the difference was statistically significant. Rats estrogen group has a high incidence of cancer.Conclusion:The BMSCs can protect perimenopausal ovarian granulosa cells in rats, BMSCs has a therapeutic effect in perimenopausal syndrome. |
