Abstract

The 2009 influenza pandemic has affected most countries of the world within a short span of time. Outbreak analysis shows that like seasonal flu great majority of the cases have mild and self-limiting illness. However, in some cases, the clinical course can deteriorate, leading to severe complications and eventually death. Research on the pathogenesis of the new virus can be of immense help in disease prevention and control.The main purpose of this study is to identify the potential differences in growth characteristics and gene expression profiles between the pandemic and seasonal A (H1N1) influenza virus while infecting A549 cells.The main contents of the study:1. Growth characteristics, including viral infection, replication, release and CPE were monitored and compared while A/California/07/09(H1N1) and seasonal influenza A/GuangdongBaoan/51/08 (H1N1) infection.2. Gene expression profiles of A549 cells were obtained using the 22 K Human Genome Oligo Array, differentially expressed genes at selected time points were screened using significance analysis of microarrays. Gene Ontology and related biological pathway analyses were performed on the differentially expressed genes with Molecule Annotation System V4.0 supported by CapitalBio(?), the KEGG database, and the OMIM database of PUBMED. The host responses to pandemic and seasonal H1N1 influenza infection were compared.The analysis results:1. We have found that both viruses can replicate effectively in A549, but differences were identified in the dynamics of the two H1N1 viruses infection. A/California/07/2009(H1N1) had a poor binding efficiency, but it could still replicate efficiently, and the viral release reached a comparable level with A/Baoan/51/2008(H1N1); The CPE due to CA/07 infection was more severe than Baoan/51.2. The immunosuppression and anti-apoptosis events of pandemic H1N1 virus were similar to those seen with seasonal H1N1 virus. Microarrays results indicated that both of the viruses suppressed host immune response related pathways including cytokine production, but pandemic H1N1 virus displayed weaker suppression of host immune response than seasonal H1N1 virus. We also observed similar anti-apoptotic events such as activation of apoptosis inhibitor and down-regulation of key genes of apoptosis pathways in both infections. The qualitative nature of the cellular response to pandemic and seasonal H1N1 infection was similar, and most of the differences were at the quantitative level. Only a few differences were identified in the alteration tendency.3. The pandemic H1N1 virus had an ability to inhibit host’s antiviral response by multiple mechanism, and could escape the innate immune defense by impeding cytokine responses, inhibiting of NK activation and evading recognition by macrophage.
